GeneGATA4-engrailed dominant negative fusion (GATA4 211-329 – engr 2-298)
Vector ConstructionMouse Gata4 cDNA for amino acids 211-329 fused to amino acids 2-298 of engrailed to make a dominant negative Gata protein. It contains 5’ flag epitope. This cassette was cloned into the multiple cloning site of pAC-CMV-pLpA. The resulting plasmid was cotransfected into HEK293 cells with plasmid pJM17 which contains the Ad5 genome. Homologous recombination between the two plasmids resulted in the replacement of the Ad5 early region 1 with the cDNA expression cassette, generating a replication deficient recombinant virus.
